Journal of Animal and Veterinary Advances, 12(14) 1198-1201, 2013 Peer-reviewedIt has been investigated that transgenic chicken can be produced by various methods by introducing an exogenous gene into chicken Primordial Germ Cells (PGCs). Blood containing PGCs was collected from the blood vessels of embryos at stages 12-15. An exogenous gene encoding Green Fluorescent Protein (GFP) was introduced into the PGCs by lipofection and electroporation. Electroporation was carried out under 200 V/25 μF, 200 V/200 μF and 200 V/950 μF. GFP expression was observed in PGCs. Efficiency of introduction into PGCs was low. Electroporation was superior to lipofection for the introduction of the exogenous gene into chicken PGCs. PGCs treated for GFP introduction by electroporation were injected into blood vessels of recipient embryos at stages 12-15. After incubation until stage 26, GFP bands were obtained from gonads of embryos, thereby demonstrating that treated PGCs had migrated to the gonad. © Medwell Journals, 2013.

JOURNAL OF POULTRY SCIENCE, 49(1) 1-4, Jan, 2012Chicken primordial germ cells (PGCs) are used to restore and conserve genetic resources of some endangered birds or flocks of domestic chickens. When PGCs are transferred from a donor to a recipient, they can survive and gain sexual maturity in the recipient chicken. This indicates that PGCs retaining exogenous genes can proliferate and develop in recipient gonads, and proves that transferred PGCs are effective sources for establishing transgenic and germline chimeric chickens. The PGC transfer method has been used to produce germline chimeric chickens and may become popular in the field of reproductive science. Germline chimeric chickens have the potential to reconstruct and reproduce endangered birds, including both domestic and wild species.

ペット栄養学会誌, 14(1) 67-68, Oct 1, 2011ビーグル犬を用いて、甘味物質であるアミノ酸と忌避物質である塩酸キニーネ(QHCl)を混合し、選択飲水をさせることによりアミノ酸の感受を行動学的に調査した。アミノ酸とQHClとの混合溶液とQHCl単独溶液では混合溶液の飲水量が増加しており、アミノ酸を感受していることが示唆された。また、アミノ酸の種類や濃度によって飲水量に相違が認められた。

Journal of Pet Animal Nutrition, 13(1) 7-11, 2010Behavioral responses of beagles to sweet taste stimuli were observed. Short-term choice tests measured the amounts water, quinine hydrochloride (QHCl) solution, and mixed solutions of QHCl and saccharin that were drunk during 20 minutes after not providing water for 12 hours. The beagles drank less QHCl solution than water (p<0.05). Solutions of 0.1 mM QHCl mixed with 1.0 or 3.0 mM saccharin were drunk less than 0.1 mM QHCl alone. There was a significant concentration-dependent decrease (p<0.05) in the drinking of solutions of QHCl mixed with sweetener. Expression levels of the T1R3 sweet receptor in dog's tongue areas (front, central, and base) were examined by real-time PCR. Expression of T1R3 mRNA was higher in the front area of the tongue than in the central and base area. Beagles seem to taste saccharin but did not like the taste in the concentrations used in this study.
