Journal of veterinary internal medicine 2023年8月18日BACKGROUND: Pulmonary hypertension (PH) secondary to respiratory disease is caused by pulmonary vascular remodeling and hypoxia. Severe PH can induce various clinical signs, including syncope and right-sided heart failure. HYPOTHESIS/OBJECTIVES: To investigate the echocardiographic characteristics in dogs with PH secondary to respiratory diseases. ANIMALS: Thirty-one dogs with respiratory diseases with or without PH and 15 healthy dogs. METHODS: Prospective cross-sectional study. Dogs were classified according to respiratory disease (obstructive airway/lung disease [OALD] or restrictive lung disease [RLD]) and PH-relevant signs. The association between echocardiographic variables and PH (classified by respiratory disease and PH-relevant signs) was investigated. RESULTS: Twenty-one dogs were diagnosed with PH; of these, 11 showed PH-related signs (OALD, n = 2; RLD, n = 9), 14 had right ventricular hypertrophy, and 19 had pulmonary arterial enlargement. Right ventricular dysfunction and dilatation were observed only in dogs with PH-related signs (n = 10). Left and right ventricular stroke volumes were significantly lower in dogs with PH (median [interquartile range]: 17.2 [12.4-20.8] and 16.8 [15.3-29.5] mL/m2 , respectively). Dogs with RLD had higher echocardiography-estimated pulmonary vascular resistance than those with OALD (median [interquartile range]: 3.1 [1.9-3.3] and 1.6 [1.3-2.2], respectively). CONCLUSIONS AND CLINICAL IMPORTANCE: Pulmonary arterial enlargement was the most common echocardiographic finding in dogs with PH secondary to respiratory diseases. Right ventricular dysfunction, dilatation, and decreased left and right ventricular stroke volume were significantly associated with the PH-related signs, indicating that comprehensive echocardiography is recommended in dogs with respiratory disease. Restricted lung disease might induce more severe PH than OALD.

Journal of veterinary internal medicine 37(3) 856-865 2023年BACKGROUND: Progression to combined post- and pre-capillary pulmonary hypertension (PH) provides prognostic information in human patients with post-capillary PH. Pulmonary vascular resistance estimated by echocardiography (PVRecho) is useful for the stratification of dogs with myxomatous mitral valve disease (MMVD) and detectable tricuspid regurgitation. OBJECTIVES: To evaluate the prognostic value of PVRecho in dogs with MMVD. ANIMALS: Fifty-four dogs with MMVD and detectable tricuspid regurgitation. METHODS: Prospective cohort study. All dogs underwent echocardiography. The PVRecho was calculated based on tricuspid regurgitation and the velocity-time integral of the pulmonary artery flow. To evaluate the influence of echocardiographic variables on cardiac-related deaths, Cox proportional hazard analysis was performed. Additionally, Kaplan-Meier curves classified by PVRecho tertiles were made and compared using log-rank tests to evaluate the influence of PVRecho on all-cause mortality and cardiac-related death. RESULTS: The median follow-up time was 579 days. Forty-one dogs with MMVD (PH severity [number]: no or mild, 21/33; moderate, 11/11; severe, 9/10) died during the study. In the multivariable Cox proportional hazard analysis adjusted for age, sildenafil administration, and American College of Veterinary Internal Medicine stage of MMVD, left atrial to aortic diameter ratio and PVRecho remained significant (adjusted hazard ratio [95% confidence interval]: 1.2 [1.1-1.3] and 2.1 [1.6-3.0], respectively). Higher PVRecho showed a significant association with lower survival rates. CONCLUSIONS AND CLINICAL IMPORTANCE: Left atrial enlargement and high PVRecho were independent prognostic factors in dogs with MMVD and detectable tricuspid regurgitation.

Veterinary Sciences 9(11) 2022年10月27日 査読有りAn 11-year-old intact female Papillion weighing 2.1 kg was referred to our institution with the main complaint of shallow, rapid breathing. At the first visit (day 0), although clinical signs improved due to the use of medication from the primary hospital, transthoracic radiography and echocardiography revealed left heart enlargement and left ventricular dysfunction. A clinical diagnosis of dilated cardiomyopathy (DCM) was made and oral administration of pimobendan, temocapril, and taurine was initiated. However, on day 10, the respiratory status worsened and furosemide was prescribed. On day 54, no significant improvement in heart size was observed. Additionally, the diet that this patient received met the recommendation for diet-related DCM by the U.S. Food and Drug Administration, and the patient's diet was changed from a grain-free diet to a grain-containing diet. On day 1191, the patient's respiratory status was stable and no clinical signs were observed. Transthoracic radiography and echocardiography revealed an improvement in left heart size. Additionally, improvements in the left and right ventricular myocardial strains were observed after changing the diet. We suggest that it may be necessary to suspect a dietary association with dilated cardiomyopathy, and a good prognosis might be expected by dietary modification, even in small-breed dogs.

Animals : an open access journal from MDPI 12(16) 2022年8月15日Pulmonary hypertension (PH) is a fatal condition that affects many dogs. In humans, PH is often treated with beraprost sodium (BPS). However, the effectiveness of BPS for canine PH has not been established. This study aimed to evaluate the clinical and cardiovascular response of BPS in canine patients with PH of various causes. Sixteen dogs with PH (post-capillary PH, n = 8; pre-capillary PH, n = 8) were included. BPS was continuously administered twice daily at 15 µg/kg. All dogs underwent echocardiography, including speckle-tracking analysis and blood pressure measurement, before and after BPS administration. Continuous BPS administration (range: 13.2-22.0 µg/kg) significantly decreased the pulmonary and systemic vascular impedance and increased left and right ventricular myocardial strain. In dogs with post-capillary PH, BPS administration caused no significant worsening of the left atrial pressure indicators. No side effects of BPS were observed in any dog. BPS also improved cardiac function and pulmonary circulation through pulmonary vasodilation, suggesting that BPS may be an additional treatment option for canine PH of various causes. Particularly, BPS increased left ventricular function and systemic circulation without worsening the left heart loading condition in dogs with post-capillary PH.

BMC veterinary research 18(1) 251-251 2022年6月28日 査読有りBACKGROUND: Cardiac tumors in cats are relatively rare, with lymphoma accounting for more than half of all cases. However, feline cardiac lymphoma is often diagnosed post-mortem, and it is difficult to diagnose while the cat is still alive. It is the first report of a direct, rather than estimative, diagnosis with cardiac needle biopsy of a living cat with cardiac lymphoma. CASE PRESENTATION: A 3-year-old domestic short-haired male cat experienced loss of energy and loss of appetite. Thoracic radiography and transthoracic echocardiography showed cardiomegaly with slight pleural effusion and cardiac tamponade due to pericardial effusion, respectively. In addition, partial hyperechoic and hypertrophy of the papillary muscle and myocardium were observed. Blood test showed an increase in cardiac troponin I levels. Pericardial fluid, removed by pericardiocentesis, was analyzed; however, the cause could not be determined. With the owner's consent, pericardiectomy performed under thoracotomy revealed a discolored myocardium. Cardiac needle biopsy was performed with a 25G needle, and a large number of large atypical lymphocytes were collected; therefore, a direct diagnosis of cardiac lymphoma was made. Pathological examination of the pericardium diagnosed at a later date revealed T-cell large cell lymphoma. The cat underwent chemotherapy followed by temporary remission but died 60 days after the diagnosis. Postmortem, two-dimensional speckle-tracking echocardiography (data when alive) revealed an abnormal left ventricular myocardial deformation, which corresponded to the site of cardiac needle biopsy. CONCLUSIONS: This rare case demonstrates that cardiac lymphoma should be added to the differential diagnosis in cats with myocardial hypertrophy and that the diagnosis can be made directly by thoracotomy and cardiac needle biopsy. In addition, the measurement of cardiac troponin I levels and local deformation analysis of the myocardium by two-dimensional speckle-tracking echocardiography may be useful in the diagnosis of cardiac tumors.

JOURNAL OF VETERINARY MEDICAL SCIENCE 58(8) 727-730 1996年8月Circulating immune complexes (CIC) in the sera from 102 clinically healthy dogs and from 16 Dirofilaria immitis (D. immitis) infected dogs were measured by the immune adherence hemagglutination (IAHA) method. The level of CIC in the sera from healthy dogs was 28.2 +/- 29.1 mu g/ml. There was no significant difference in the levels of CIC regarding sexes or ages. On the other hand, the level of CIC in the sera from D. immitis infected dogs was 230.3 +/- 117.1 mu g/ml, which was significantly (p<0.01) higher than that of healthy dogs. The results of this study confirm that IAHA method is one of the reliable means of detecting and/or diagnosing immune complex mediated diseases in dogs.

JOURNAL OF VETERINARY MEDICAL SCIENCE 58(5) 451-453 1996年5月Cardiac arrest occurred in a male Labrador Retriever dog weighing 27.8 kg during induction to anesthesia. Immediately after the failure of resuscitation by the external cardiac compression, thoracotomy was performed and open chest direct current (DC) counter shocks were applied with routine emergency medications. Then the dog recovered consciousness. Although cardiac rhythm just after resuscitation was sinus tachycardia with paroxysmal supraventricular tachycardia, multifocal ventricular arrhythmia occurred 2 hr after resuscitation. This arrhythmia might be the result from reversible cardiac lesions due to DC counter shock.

The journal of veterinary medical science 58(4) 381-384 1996年4月両房室弁閉鎖不全による重度のうっ血性右心不全の犬2例を経験した. 2例とも老齢の雄小型犬であった. うっ血性心不全の原因は主に三尖弁閉鎖不全で, 発咳などの左心不全症状はほとんど見られなかった. 三尖弁閉鎖不全は続発性ではなく, 原発性であると考えられた.

JOURNAL OF BIOLOGICAL CHEMISTRY 269(37) 23274-23279 1994年9月We have cloned a second class of inward rectifier potassium channels, designated MB-IRK2, from a mouse brain cDNA library. The amino acid sequence of this clone shares 70% identity with the mouse IRIK1. Xenopus oocytes injected with cRNA derived from MB-IRIK2 expressed a K+ current, which showed inward rectifying channel characteristics similar to the MB-IRK1 current. In contrast to the MB-IRK1 current, however, the MB-IRK2 current exhibited significant inactivation during hyperpolarizing pulses. In patch clamp experiments with 140 mM K+ in the pipette, the single channel conductance of MB-IRK2 was 34.2 +/- 2.1 picosiemens (n = 5), a value significantly larger than that of MB-IRK1 (22.2 +/- 3.0 picosiemens, n = 5). Consistent with the whole cell current, the steady-state open probability (P-o) of the MB-IRK2 channel decreased with hyperpolarization, whereas that of the MB-IRK1 remained constant. Northern blot analysis revealed the mRNA for MB-IRK2 to be expressed in forebrain, cerebellum, heart, kidney, and skeletal muscle. In the brain, the abundance of mRNA for MB-IRK2 was much higher in cerebellum than in forebrain and vice versa in the case of MB-IRK1. These results demonstrate that the IRK family is composed of multiple genes, which may play heterogenous functional roles in various organs, including the central nervous system.

FEBS LETTERS 346(2-3) 251-256 1994年6月We have cloned a novel brain-specific inward rectifier K+ channel from a mouse brain cDNA library and designated it MB-IRK3. The mouse brain cDNA library was screened using a fragment of the mouse macrophage inward rectifier K+ channel (IRK1) cDNA as a probe. The amino acid sequence of MB-IRK3 shares 61% and 64% identity to MB-IRK1 and RB-IRK2, respectively. Xenopus oocytes injected with cRNA derived from this clone expressed a potassium current which showed inward-rectifying channel characteristics similar to MB-IRK1 and RB-IRK2 currents, but distinct from ROMK1 or GIRK1 current. However, the single channel conductance of MB-IRK3 was similar to 10 pS with 140 mM extracellular K+, which was distinct from that of MB-IRK1 (20 pS). MB-IRK3 mRNA expressed specifically in the forebrain, which clearly differed from MB-IRK1 and RB-IRK2 mRNAs. These results indicate that members of the IRK family with distinct electrophysiological properties express differentially and may play heterogenous functional roles in brain functions.

The rat brain cDNA library was screened using a fragment of the mouse macrophage IRK1 cDNA as a probe. The amino acid sequence of RB-IRK2 shares 70%, 40% and 45% identity to mouse IRK1, rat ROMK1 and rat GIRK1, respectively. Xenopus oocytes injected with cRNA derived from RB-IRK2 expressed a potassium current which showed inward-rectifying channel characteristics similar to the IRK1 current, but distinct from the ROMK1 or the GIRK1 currents. However, the localization of RB-IRK2 mRNA in rat tissues, assessed by the Northern blot analysis, differed from that of mouse IRK1. These results indicate that the IRK family is composed of multiple genes, which express in different tissues and therefore may play heterogenous functional roles in various organs, including rat central nervous system. © 1994.

Research in Veterinary Science 57 251-252 1994年1月1日The concentration and molecular form of the plasma atrial natriuretic peptide (ANP) in normal calves during the first 10 days of life were investigated. The mean (SEM) ANP concentration on the day of birth was 62·7 (5·52) pmol litre-1, and thereafter it decreased progressively and significantly (P<0·001). After 10 days, the plasma ANP concentration had decreased to normal adult values (11·3 [3·87] pmol litre-1). On the day of birth and 10 days later a single peak corresponding to α-ANP alone was detected in the plasma of the calves. © 1994.

東北家畜臨床研究会誌 17(1) 20-24 1994年乳房炎罹患牛55頭76分房に対し泌乳期用セファピリン注入剤(KP-10)の治療試験を実施した。KP-10の投与は1日1回、1分房に1本、3日間連続して注入した。対照薬剤はベンジルペニシリン、硫酸ジヒドロストレプトマイシン配合剤を同様に注入した。効果の判定は、薬剤注入前と注入後3~5日目における臨床検査、乳汁検査及び細菌検査により行った。その結果臨床所見、細胞数および細菌数の総合判定による有効率はKP-10が85.4%対照薬剤が64.3%であった。(P<0.05)。<BR>臨床所見における、有効率はKP-10が72.9%、対照薬剤は57.1%であった。細胞数の改善率はKP-10が97.7%、対照薬剤が75.0%であった(P<0.01)。乳汁の中の起炎菌の陰性化率はKP-10が83.3%、対照薬剤が57.1%であった(P<0.02)。起炎菌別の有効率はKP-10についてはブドウ球菌が89.5%、レンサ球菌が62.5%であった。投薬前の乳汁から分離された起炎菌はセファピリンに対して強い感受性を示したが、ペニシリン及びストレプトマイシンに感受性.が低下した株も見られた。以上の結果から、KP-10は泌乳期乳房炎に対して有効であることが認められた。

日本獣医師会雑誌 47(9) 655-657 1994年成長不良, 短尾 (3cm) および腹水を呈した3ヵ月齢雌の38kgのホルスタインの子牛で, 心室中隔欠損および騎乗大動脈が認められた. 脊椎後列では, S3以降で右棘突起の低形成, L6とC1間の二分脊椎が見られた.尾椎は2個の骨からなり, 最後尾椎は融合した椎骨であった.

FEBS LETTERS 336(3) 375-380 1993年12月We have cloned an inward-rectifier potassium channel from a mouse brain cDNA library, studied its distribution in the brain by in situ hybridization and determined the chromosomal localization of the gene. A mouse brain cDNA library was screened using a fragment of the mouse macrophage IRK1 cDNA as a probe. Two duplicate clones of similar to 5.5 kb were obtained. Xenopus ococytes injected with cRNA derived from the clone expressed a potassium channel with inwardly rectifying channel characteristics. The amino acid sequence of the clone was identical to that of IRK1 recently cloned from a mouse macrophage cell line. In situ hybridization study showed the mouse brain IRK1 to be generally distributed throughout the brain, but in particular subsets of neurons at high levels. The gene was placed in the distal region of mouse chromosome 11, which contains several uncloned neurological mutations. These results provide the first demonstration of the cloning and distribution of an inward rectifier potassium channel from the nervous system.

日本獣医師会雑誌 46(12) 1037-1040 1993年1ヵ月来呼吸促迫と腹水貯留を示した猫 (ヒマラヤン種, 雌, 8歳) に, ジギタリス剤, 利尿剤および血管拡張剤による治療を行ったところ, うっ血性心不全症状は改善されたが, 腎不全の悪化により第52病日に死亡し, 剖検で拡張型心筋症が確認された.

The journal of veterinary medical science 54(4) 789-790 1992年8月陽イオン交換カラムを使用した高速液体クロマトグラフィー(HPLC)によりネコ糖化ヘモグロビンを測定した. 臨床上正常な猫37頭及び糖尿病猫12頭の糖化ヘモグロビンの百分比はHbAlcではそれぞれ1.70%および3.54%, HbAlでは1.88%および3.85%と, 糖尿病猫で有意な高値を示していた(Plt;0.05). このことからネコ糖化ヘモグロビンは糖尿病診断の指標になると思われた.

日本獣医師会雑誌 45(2) 106-108 1992年心房細動を併発した右心不全犬における血漿中の心房性ナトリウム利尿ペプチド (ANP) の濃度および分子型について検討した. 血漿中ANP濃度は119.4±40.3pg/m<I>l</I> (n=5) で, 健常犬 (43.5±1.86pg/m<I>l</I>, n=20) と比較して有意に高値を示した (p<0.05). また, 血漿中ANPの分子型は健常犬で認められるα-ANP (MW3,000) に加えて, 高分子型であるβ-ANP (MW6,000) やγ-ANP (MW13,000) が検出された.

日本獣医学雑誌 53(1) 65-68 1991年2月15日我々は陽イオン交換カラムを使用した高速液体クロマトグラフィー (以下HPLC) による, イヌ糖化ヘモグロビンの測定法を確立した. イヌヘモグロビンは, 2つのmajorピーク (HbA0) と, HbA1a, HbA1b, HbA1cからなる3つのminorピークに分画された. 臨床的に正常な犬38頭と糖尿病犬10頭でのHbA1cの平均値はそれぞれ2.60および6.41%であった. またHbA1の平均値はそれぞれ3.58および7.41%となり, 糖尿病犬のHbA1c及びHbA1は正常犬に比較して有意な高値を示した (p<0.01). イヌ糖化ヘモグロビンの測定におけるHPLC法の有用性は高く, インスリン療法のモニターへの臨床応用が可能であろうと考えられた.
