International Journal of Molecular Sciences 25(11) 2024年6月An angiotensin receptor/neprilysin inhibitor (ARNI), a heart failure treatment, is a combination drug made up of sacubitril, a neprilysin inhibitor, and valsartan, a vascular receptor blocker. No human or veterinary studies regarding the effect of ARNI on renal haemodynamics in the absence of cardiac or renal issues exist. Therefore, we investigated the effect of ARNI on renal haemodynamics in five healthy dogs. ARNI was administered to all five dogs at an oral dose of 20 mg/kg twice daily for 4 weeks. Renal haemodynamics were assessed on the day before ARNI administration (BL), on Day 7, and on Day 28. The glomerular filtration rate (GFR) significantly increased on Day 28 compared to BL and Day 7, whereas renal plasma flow increased on Day 7 and Day 28 compared to BL. Systolic blood pressure significantly decreased between BL and Day 28. Plasma atrial natriuretic peptide (ANP) concentrations increased on Day 7 compared to BL. Additionally, ANP concentrations increased on Day 28 in three of the five dogs. Different ANP concentrations were observed in the remaining two dogs. Both urine output volume and heart rate remained relatively stable and did not exhibit significant change. In conclusion, ARNI may enhance renal haemodynamics in healthy dogs. ARNI could be a valuable drug for treating both heart and kidney disease in dogs.

Open Veterinary Journal 13(5) 604-604 2023年Background: Plasma N-terminal pro-atrial natriuretic peptide (NT-proANP) and plasma N-terminal pro-B-type natriuretic peptide (NT-proBNP) concentrations may be affected by the hydration status.Aim: This study aimed to evaluate the effect of dehydration on plasma NT-proANP and NT-proBNP concentrations in healthy dogs. Methods: This prospective study included five clinically healthy dogs. Furosemide was administered intravenously at 2–4 mg/kg every 1–2 h until completion of the dehydration model. The dehydration model was considered complete when weight loss was ≥5% and findings of dehydration on physical examination were observed. Plasma NT-proANP and NT-proBNP concentrations were compared at three time points: before the dehydration model was created (point 1), at the completion of the dehydration model (point 2), and when dehydration was judged to have improved (point 3). Association between plasma NT-proANP and NT-proBNP concentrations, and each clinical variable (physical examination, blood pressure, blood chemistry, blood gases, and echocardiography) was assessed using linear regression analysis. Results: Plasma NT-proANP concentration decreased significantly from point 2 to point 1 (p < 0.05), whereas plasma NT-proBNP concentration showed a decreasing trend but did not differ significantly between points 1 and 2. Plasma NT-proANP concentration correlated significantly with body weight (R2 = 0.178) and plasma NT-proBNP concentration (R2 = 0.284) (p < 0.05, respectively), and plasma NT-proBNP concentration correlated significantly with electrolytes (sodium, R2 = 0.439; potassium, R2 = 0.444; chloride, R2 = 0.419), and echocardiographic parameters (diastolic left ventricular internal diameter [LVIDd], R2 = 0.519; weight-standardised LVIDd, R2 = 0.535) (p < 0.01, respectively). Conclusion: The plasma NT-proANP concentrations decreased with dehydration. However, the plasma NT-proBNP concentration did not change with mild dehydration and reflected left ventricular morphology.

Veterinary record open 9(1) e28 2022年12月BACKGROUND: Ivabradine is used to treat tachycardia; unlike atenolol, it does not affect blood pressure or myocardial contractility. This study compared the impact of ivabradine and atenolol on heart rate (HR) and HR variability (HRV) during a 24 h period, feeding and sleeping times, via a Holter electrocardiogram in healthy cats. We hypothesised that ivabradine and atenolol would lower the HRs equally well, even at times of excitement and rest, such as during feeding and sleep; that ivabradine, unlike atenolol, would have an effect on HRV. METHODS: Five clinically healthy cats were used in the prospective blinded crossover study receiving 3 days of ivabradine (0.30 mg/kg per os twice daily) followed by atenolol (6.25 mg/cat per os twice daily, range 1.3-2.0 mg/kg) or receiving atenolol followed by ivabradine. A placebo period was initiated before the start of the crossover test, data obtained during that period were used as a baseline (BL). Evaluation parameters included HR and HRV, for the whole 24 h period and for feeding and sleeping times, comparing the effect of ivabradine and atenolol with BL. RESULTS: The HR for the whole 24 h, feeding and sleeping times, were significantly lower with ivabradine and atenolol, compared to BL (p < 0.05). The HRV for the whole 24 h and sleeping time were significantly higher after ivabradine compared with BL and after atenolol. CONCLUSIONS: In healthy cats, ivabradine and atenolol significantly reduced the HR regardless of excitement and rest; their effects were comparable. Ivabradine significantly increased HRV in comparison to BL whereas atenolol did not.

Veterinary World 2072-2083 2022年8月27日Background and Aim: Electrocardiography (ECG) is an adjunct for cardiac enlargement diagnosis. However, its efficacy in assessing left cardiac remodeling (left atrial and left ventricular enlargement) in dogs with myxomatous mitral valve disease (MMVD) remains unclear. This study aimed to evaluate the association between ECG parameters and left cardiac remodeling and to investigate whether the rate of change in ECG waveforms in the same individual reflected left cardiac remodeling in dogs with MMVD.Materials and Methods: This retrospective study included 20 healthy dogs and 140 dogs with MMVD. Data on clinical variables were obtained through physical examination, thoracic radiography, and echocardiography. The ECG parameters were the P-wave duration, PR interval, QRS complex duration, P-wave amplitude, R-wave amplitude, and mean electrical axis. Dogs with examination data that could be obtained multiple times during the study period were classified into the non-progressive and progressive groups.Results: Only the P-wave and QRS complex durations were selected as significant variables associated with imaging test parameters (p < 0.05); they had a relatively higher discriminatory ability for the left cardiac remodeling than other ECG parameters. The rates of change in the PR interval and R-wave amplitude were significantly higher in the progressive group than in the non-progressive group.Conclusion: In dogs with MMVD, the P-wave and QRS complex durations were significantly correlated with the left cardiac remodeling indicators. Furthermore, an increased rate of change in the PR interval and R-wave amplitude in the same individual may indicate left cardiac remodeling.

The journal of veterinary medical science 61(4) 421-423 1999年4月11ヵ月齢の雌の黒毛和種に鼻腔ノカルジア症がみられた. 鼻腔の左側後部は黄褐色乾酪物質によって完全に閉塞し, また脳の嗅球および前頭葉に少数の褐色軟化単が認められた. 組織学的には肉眼病変に一致して, 細胞残骸物の中心部とそれを取り巻く類上皮細胞, 好中球, リンパ球およびラングハンス型多核巨細胞からなる肉芽腫が密発していた. 病巣内には糸状で分技を示すグラム陽性, かつ一部抗酸性の桿菌が多数認められた.

JOURNAL OF SMALL ANIMAL PRACTICE 39(12) 592-594 1998年12月A two-year-old intact male shiba inu dog with excessive polyuria and polydipsia (PU-PD) was diagnosed as having congenital nephrogenic diabetes insipidus based on clinical findings, the results of urinalysis, blood examinations, a modified water deprivation test and a low dose dexamethasone suppression test. The owner was advised to provide adequate access to drinking water, and treatment with a low dose of hydrochlorothiazide (2 mg/kg, twice daily) together with a low sodium diet was initiated. As a result, the daily water intake decreased significantly from 6500 to 7500 ml/day (800 to 980 ml/kg/day) to 1400 to 1900 ml/day (170 to 230 ml/kg/day) and the clinical signs associated with the PU-PD and dehydration improved over the following two years.

Laboratory Animal Science 48(4) 353-356 1998年A new experimental rat model for suppression of release of arginine vasopressin (AVP) was evaluated. Release of AVP is potentiated by physiologic stimuli, but is suppressed by emotional stress. Rats were exposed to the aggressive behavior of other rats injected with 6-hydroxydopamine hydrochloride (6-OHDA) in both lateral ventricles and subjected to a pain stimulus applied to the tail. To ascertain whether emotional stress is induced by use of this method, plasma corticosterone, glucose, and AVP concentrations were measured in the stressed group of rats (n = 8) placed near a group of aggressive (fighting) rats. Characteristic changes in behavior, significant increases in plasma corticosterone and glucose values, and lack of increase in plasma AVP concentration in stressed rats confirmed that they experienced emotional stress. This new model meets the experimental requirements for suppression of AVP release in rats.

日本獣医師会雑誌 51(3) 141-143 1998年ホルスタイン種雌牛 (5歳) の喉頭から頸部腹側皮下に波動感を呈する腫瘤 (径20cm) がみられ, 血清T<SUB>4</SUB>およびT<SUB>3</SUB>濃度は高値を示したが, 腫瘤内容の血様漿液から細菌および腫瘍細胞は検出されなかった. 合併した迷走神経性消化不良と腎不全の対症療法を実施したが死亡し, 腫瘤では病理組織学的に濾胞を形成しつつ増殖する腫瘍性腺細胞とともに, 周辺組織への顕著な浸潤増殖がみられ, 甲状腺癌と診断された.

動物の循環器 31(2) 59-68 1998年小動物の慢性心不全療法においてACEIは不動の地位にある。これまでに報告されている知見は,診察現場にとって貴重な情報ではあるが,即座に臨床例に外挿できない側面が残されていることを私見として述べたい。また,著者の未公表データに基づき,心不全初期からのACEIの投与が病態の進行を遅延させる可能性があることを紹介した。さらに,ACEI投与中高窒素血症に関しても若干の考察を加えた。

JOURNAL OF VETERINARY MEDICAL SCIENCE 59(11) 1067-1069 1997年11月A fluorometric assay for the determination of hepatic carnitine palmitoyltransferase (CPT) activity was slightly modified for use with cattle samples. With this assay, the K-m value was 0.56 +/- 0.10 mM with respect to L-carnitine (mean +/- SD, n=4) and was 9.6 +/- 2.2 mu M (n=3) with respect to palmitoyl-CoA. The average hepatic CPT activity was 33.6 +/- 2.0 mu mol CoASH/min/g protein in 38 healthy cattle and was similar in both sexes and among breeds. Hepatic CPT activity showed no correlation with serum phospholipid, free fatty acid, triglyceride or total cholesterol concentrations.

The journal of veterinary medical science 59(11) 1067-1069 1997年11月蛍光法を用いたCarnitine Palmitoyltransferase (CPT) 活性を牛用に応用した. 本法によるK_m値はL-carnitineに対して0.56±0.10mM (mean±SD, n=4), palmitoyl-CoAに対して9.6±2.2μM (n=3) であった. 健康牛38頭における平均CPT活性は33.6±2.0μmol CoASH/min/g proteinで, 性および品種間に差はなく, また血清リン脂質, 遊離脂肪酸, トリグリセリドおよびコレステロール値と相関はなかった.

JOURNAL OF VETERINARY MEDICAL SCIENCE 59(5) 395-396 1997年5月Circulating immune complexes (CIC) in the sera from 45 clinically healthy cats and 23 feline immunodeficiency virus (FIV) infected cats were measured by an immune adherence hemagglutination (IAHA) method. The level of CIC in the sera from FIV sera-negative healthy cats was 47.2 +/- 47.3 mu g/ml when expressed as heat aggregated feline IgG equivalent value in IAHA reactivity. On the other hand, the level of CIC in the sera from FIV infected cats was 757.4 +/- 910.5 mu g/ml, which was significantly higher than that of healthy ones. CIC levels of 11 symptomatic cats and 12 asymptomatic ones were 837.8 +/- 1138.2 mu g/ml and 683.0 +/- 684.2 mu g/ml, respectively. These results showed that IAHA method was reliable to detect CIC levels of cat sera and that CIC levels in the sera of cats infected with FIV were higher than those of healthy ones.

The journal of veterinary medical science 59(5) 395-396 1997年5月ネコ免疫不全ウイルス(FIV)抗体の認められない45頭の健常ネコを対照として23頭のFIV感染ネコの血中免疫複合体(CIC)を, 免疫粘着赤血球凝集反応法(IAHA)によって測定した. FIV感染ネコのCIC値(757.4±910.5μg/ml)は, 健常ネコ(47.2±47.3μg/mlに比して有意に高かった. また, 臨床症状を示すFIV感染ネコ(11頭)と臨床症状を示さないFIV感染ネコ(12頭)のCIC値はそれぞれ, 837.8±1138.2μg/mlと683.0±684.2μg/mlであった. 以上の結果より, IAHA法によってネコのCIC血中濃度は測定可能で, FIV感染ネコでは高値を示すことが判明した.

LABORATORY ANIMAL SCIENCE 47(2) 190-193 1997年4月A new technique for collecting blood from conscious and unrestrained rats is reported, Plasma arginine vasopressin (AVP) concentration has been known to be influenced by various procedures used for blood collection (e.g., manual contact and anesthesia), In the study reported here, the improvement of the routine jugular vein cannula method was undertaken by extending the connection tubing to allow use of a swivel mechanism, Plasma AVP, glucose, and corticosterone concentrations in virgin rats were measured in blood samples collected by use of the new method and of two generally accepted methods (decapitation and routine jugular vein cannula methods), All values of the three parameters obtained were the lowest when the new method was used, suggesting that this new swivel method induced the least stress in the rats.

The journal of veterinary medical science 58(8) 727-730 1996年8月臨床的に健常でDirofilaria immitis (D.immitis)感染の認められない102頭のビーグルと16頭のD.immitis感染犬の血中免疫複合体(CIC)を免疫粘着赤血球凝集反応(IAHA)法によって測定した. 102頭の健常犬のCIC値は, 28.2±29.1μg/mlであった. CIC値に関しては, 年齢や性による有意差はみられなかった. D.immitis感染犬のCIC値(210±111.4μg/ml)は, 健常犬に比べ有意に高かった(P<0.01). 今回の実験結果より, IAHA法がイヌのCIC測定に適した方法であり免疫複合体介在性疾患の検出あるいは診断に応用可能であると考えられた.

JOURNAL OF VETERINARY MEDICAL SCIENCE 58(8) 727-730 1996年8月Circulating immune complexes (CIC) in the sera from 102 clinically healthy dogs and from 16 Dirofilaria immitis (D. immitis) infected dogs were measured by the immune adherence hemagglutination (IAHA) method. The level of CIC in the sera from healthy dogs was 28.2 +/- 29.1 mu g/ml. There was no significant difference in the levels of CIC regarding sexes or ages. On the other hand, the level of CIC in the sera from D. immitis infected dogs was 230.3 +/- 117.1 mu g/ml, which was significantly (p<0.01) higher than that of healthy dogs. The results of this study confirm that IAHA method is one of the reliable means of detecting and/or diagnosing immune complex mediated diseases in dogs.

The journal of veterinary medical science 58(5) 451-453 1996年5月麻酔導入時に発生したイヌの心停止を開胸下DCカウンターショックで救命した. 蘇生直後の心調律は発作性上室性頻脈が間入する洞性頻脈が主体であったが, 2時間後には多源性心室性頻脈に移行した. この不整脈は, DCカウンターショックによる可逆的心筋障害から生じたと考えられた.

JOURNAL OF VETERINARY MEDICAL SCIENCE 58(5) 451-453 1996年5月Cardiac arrest occurred in a male Labrador Retriever dog weighing 27.8 kg during induction to anesthesia. Immediately after the failure of resuscitation by the external cardiac compression, thoracotomy was performed and open chest direct current (DC) counter shocks were applied with routine emergency medications. Then the dog recovered consciousness. Although cardiac rhythm just after resuscitation was sinus tachycardia with paroxysmal supraventricular tachycardia, multifocal ventricular arrhythmia occurred 2 hr after resuscitation. This arrhythmia might be the result from reversible cardiac lesions due to DC counter shock.

The journal of veterinary medical science 58(4) 381-384 1996年4月両房室弁閉鎖不全による重度のうっ血性右心不全の犬2例を経験した. 2例とも老齢の雄小型犬であった. うっ血性心不全の原因は主に三尖弁閉鎖不全で, 発咳などの左心不全症状はほとんど見られなかった. 三尖弁閉鎖不全は続発性ではなく, 原発性であると考えられた.

JOURNAL OF VETERINARY MEDICAL SCIENCE 58(4) 381-384 1996年4月Congestive heart failure resulted from bilateral atrioventricular insufficiency was diagnosed in two, small breed, intact male aged dogs. In both cases, the clinical signs were mainly associated with right heart failure, while those related to left heart failure were seldom observed. Radiographic and echocardiographic examinations revealed the dilatation of the right heart due to tricuspid valve insufficiency. Mitral regurgitation was also observed without pulmonary hypertension. Both patients responded to medications of diuretics, methyldigoxin and angiotensin converting enzyme inhibitor. As congestive signs deteriorated, they became unresponsive to the treatment. Ultimately, the dogs died 9 and 13 months later from the first admission.
