    Most testicular germ cell tumors have serological tumor markers such as alpha-fetoprotein (AFP) and human chorionic gonadotropin beta-subunit (beta-HCG). On the other hand, molecular staging of these tumors has not been well established compared to other urogenital malignancies like prostate cancer. Recently, melanoma antigen (MAGE) which is one of the tumor-associated antigens recognized by cytotoxic T lymphocytes (CTL) has been found to be present in a variety of malignant tumor types and normal testis. In addition, Wilms' tumor-associated gene WT1 has been found to be expressed in some testicular cancers. Thus, we examined the expression of these genes in testicular cancer tissues and peripheral blood of cancer-bearing patients using reverse transcription-polymerase chain reaction (RT-PCR). The expression of the MAGE1-3 genes was examined in 34 testicular germ cell tumors (24 seminomas and 10 nonseminomas). Of the seminomas and nonseminomas, 87.5% and 40% were positive for MAGE1, 91.7% and 60% for MAGE2, and 83.3% and 30% for MAGE3, respectively. The expression of the MAGE genes was not correlated with the tumor stage. The expression of the WT1 gene was quantified in 26 testicular germ cell tumors. WT1 was highly expressed in 5 of the 7 high stage cases, but in only 4 of the 19 low stage cases (p < 0.01). The mRNA of these genes could not be detected from the peripheral blood of patients with high stage tumors. These results suggest that MAGE genes may be useful tumor markers for molecular staging of testicular cancer, especially seminoma, and that the WT1 gene may be a tumor marker for high stage testicular germ cell tumors. However, these genes cannot yet be used for molecular staging of testicular germ cell tumors.

    A method for refining grains near the surfaces of metal workpieces using hot shot peening is proposed to improve the surface properties of the workpiece. In this method, the grains are refined due to plastic deformation generated by the hit of many shots under hot working conditions. A model experiment using two shots is carried out to examine the effects of the amount of deformation, the processing temperature and the interval of the hit on the grain size. In the experiment, the workpieces are stainless steel SUS304, carbon steel S25C, mild steel and commercially pure copper. By means of peening with many shots, the global surface layer successfully attains to the fine grains.

    Objectives: To evaluate the natural history and the efficacy of treatments for renal cell carcinoma (RCC) with brain metastasis, we reviewed 18 patients with this disease. Methods: Out of 325 cases with RCC treated at Osaka University Hospital from 1957 to 1993, 18 15.5%, male:female ratio 16:2) cases developed brain metastases. Median follow-up was 44 months after the initial treatment of the primary lesion. Twelve patients had surgical resection of brain metastases (surgical group), and 7 of them received adjuvant radiotherapy. Six patients with poor performance status were treated with supportive therapy alone (nonsurgical group). Results: Of 18 RCC patients with brain metastasis, 16 were male and 2 female. All brain metastases except for 1 case were symptomatic. Median interval between the initial treatment of the primary lesion and the diagnosis of brain metastasis was 19 months. The most frequent metastatic site prior to brain was the lung, which was detected in 7 cases (38.9%). Median survival of the entire group, measured from the onset of brain metastasis, was 9.5 months. One-year survival rate after the diagnosis of brain metastasis was 43.2% (64.8% in surgical group, 0% in nonsurgical group), 3-year 18.5% and 5-year 0%. Among 109 metastatic RCC, 14 patients were treated by lymphokine-activated killer (LAK) therapy. Out of 14 metastatic RCC patients treated by LAK therapy, 3 (21.4%) developed brain metastases. On the other hand, out of 95 metastatic RCC patients without LAK therapy, 15 (15.8%) had brain metastases. There was no significant difference in the rate of brain metastases between these two groups. Conclusion: There was a trend for prognosis of the surgical group to be better compared to that of the nonsurgical group, although it is not statistically significant. The optimum treatment for brain metastasis of RCC remains undefined, but our data suggested surgical resection in selected patients might contribute to prolonged survival of patients with brain metastasis. LAK therapy was not necessarily the risk factor of the brain metastasis.

    A 55-year-old male visited our hospital with a complaint of gross hematuria and right lower abdominal pain. Cytological findings of voided urine suggested the presence of malignant cells. Cystoscopic examination revealed bloody urine discharge from the right ureteral orifice and no abnormality in the bladder wall. The retrograde pyelogram showed no tumor masses. However, malignant cells were detected cytologically in the right ureteral catheteral urine twice. Under the preoperative diagnosis of primary urothelial tumor of the right upper urinary tract, right total nephroureterectomy was performed. A histological study revealed transitional cell carcinoma in situ in the lower portion of the ureter. We reviewed 46 cases of primary carcinoma in situ of the upper urinary tract previously reported in Japan.

    Shot peening, one of the surface treatment method, has widely applied to exert a positive influence on fatigue by the plastic deformation and the compressive residual stress of surface layers.     A method for lining workpieces with dissimilar foils using shot peening is proposed. The foil is welded to the surface of the workpiece due to plastic deformation caused by the hit of many shots. To examine the possibility of welding, point welding using a single shot is performed, and the bond strength is evaluated by tearing the foil from the workpiece. In this experiment, the workpieces are mild steel, commercially pure copper and aluminium, and the foils are commercially pure aluminium and mild steel of 0.1 mm thickness. The lining of the soft metal with the hard metal and the reverse combination are dealt with. The weldability is also heightened by heating the foil and workpiece. By means of peening with many shots, the aluminium foil is successfully welded over the surface of the mild steel workpiece. © 1998 Elsevier Science S.A. All rights reserved.

    A 63-year-old male visited our hospital with a complaint of painless swelling of the left scrotum. Left high orchiectomy was performed since ultrasonography suggested a testicular tumor. Histologically, this testicular mass was a Leydig cell tumor. We reviewed 47 cases of this tumor previously reported in Japan.

    (Purpose) To evaluate the natural history and the efficacy of treatments for carcinoma in situ (CIS) of the urinary bladder, we reviewed 70 patients with this disease. (Method) The patients of CIS were divided into 3 groups based on tumor history: primary, secondary and concurrent CIS. We studied 70 patients with primary and secondary CIS treated at our hospital between 1957 and 1995, exclusive of concurrent one. They included 31 patients with primary CIS and 39 with secondary one, ranging in age from 39 to 82, with the average age of 63.1 years. Sixty-one patients were male and 9 were female. (Results) In 31 patients with primary CIS, 12 (32.8%) complained of gross hematuria, 10 (32.3%) urinary frequency and 9 (29.0%) pain on urination. The 5-year survival rates for primary and secondary CIS were 89.1% and 91.4%, and the 5- year bladder preservation rates were 54.5% and 57.6%, respectively. Three of 31 patients with primary CIS developed an invasive carcinoma, whereas 4 of 39 patients with secondary CIS did. There was no significant difference between each primary and secondary groups. In both primary and secondary CIS, the group of immediate total cystectomy was not significantly differ from the groups of the following total cystectomy and the bladder preservation in the effect of total cystectomy on the survival rate. A total of 51 cases of CIS was treated with intravesical therapy, 17 intravesical Bacillus Calmette- Guerin (BCG) therapy, 34 intravesical chemotherapy, respectively. Intravesical BCG therapy has shown the complete response rate of 78.6%, intravesical chemotherapy 42.1%. And the bladder preservation rate seemed to be better (but not significantly) in intravesical BCG group than in intravesical chemotherapy group or non-treatment group of intravesical therapy. (Conclusion) For both primary and secondary CIS, the 5-year survival rate was about 90% and 5-year bladder preservation rate was over 50%. There was no significant difference between the primary and secondary group. The group of immediate total cystectomy was not significantly differ from the groups of the following total cystectomy and the bladder preservation in the effect of total cystectomy on the survival rate. About intravesical therapy, BCG was very effective in CR rate and the bladder preservation rate seemed to be better (but not significantly) in BCG group than in chemotherapy or non- treatment group.

    Chromium-nickel sintered compacts containing 50 and 80 mass%Cr were tried to improve their mechanical properties by means of a thermo-mechanical treatment, i.e., repeated rolling and annealing. Specimens from the as-sintered compacts showed no ductility at room temperature and small ductility at temperatures higher than 1000 K because of poor alloying of nickel with chromium. Tensile properties of the sheets repeatedly rolled with the intermediate annealing at 1173 K were very similar to those of the as-sintered specimen. While the sheets rolled with the annealing at 1573 K were excellently improved in their strength and ductility due to the considerable interdiffusion of chromium and nickel. The intermediate temperature embrittlement, that is, the ductility minimum well observed around 1000 K in a chromium-nickel alloy appeared also in the sheets rolled with the annealing at 1573 K.

    Purpose: Inhibitory effects of TNP-470, a synthetic analogue of the antibiotic fumagillin secreted by Aspergillus fumigatus, were studied with respect to growth and lung metastasis of the hormone-independent rat prostatic carcinoma cell line AT6.3. Materials and Methods: Rat prostatic carcinoma AT6.3 cells were implanted in nude mice subcutaneously. Antimetastatic and growth-inhibitory effects of TNP-470 in vivo were examined 3 weeks after inoculation of AT6.3 cells. Direct growth-inhibitory effect was examined by MTT assay in vitro. Results: TNP-470 inhibited the growth of AT6.3 cells in vitro. Subcutaneously injected TNP-470 markedly reduced numbers and individual size of lung metastases from AT6.3 cells inoculated percutaneously or intravenously into male BALB/c-nu/nu mice. Conclusion: This agent, which acts as an angiogenesis inhibitor, may prove to be clinically useful in preventing metastasis of hormone-independent prostatic cancer.

    The present paper describes how the cladding of chromium plate with dissimilar metals improves the plastic bendability of the chromium. Three-point bending tests at various temperatures were performed for three types of chromium specimens: a monolithic chromium plate, aluminium- and steel-clad chromium plate. The aluminium-clad chromium plate was bent at 343 K up to a bent angle of 90 degrees without failure, even when the chromium layer was located outside of the plate (tension side), while the monolithic chromium plate could be bent exclusively at temperatures above 403 K. When the chromium layer was located inside of the steel-clad chromium plate (compression side), the plate was successfully bent at 307 K. The FE stress analysis of bending proved that the cladding of chromium plates with proper metals of different kinds is effective to reduce the tensile stress in chromium induced during bending and also the residual stress existing after bending operation.

    The formability of sintered powder chromium plates of 99.8%-purity in roll-compression bending is investigated at various temperatures below the DBTT (Ductile-to-Brittle Transision Tempearture: 180°C for the chromium). In the roll-compression bending method for less-ductile chromium, roll-contact pressure is expected to prevent surface cracking, because high hydrostatic pressure increases ductility. However, on the first trial for monolithic chromium plates, a problem of this method was found from both the experiments and the FEM stress analysis that high tensile stress appears near the roll-contact region of the plate. To reduce the tensile stress there, steel-covered chromium plates were tested and were successfully bent at 80°C, whereas three-point bending was applicable only at the temperature above 130°C. It is thus confirmed that the roll-compression bending of steel-covered chromium plates is quite effective in decreasing the forming temperature.

    OBJECTIVES: TNP-470, a synthetic analogue of fumagillin which is a naturally secreted antibiotic produced by Aspergillus fumigatus, has reportedly antiangiogenic activity. However, there has been no report on its efficacy against metastasis of prostatic cancer. In this study, we have evaluated the inhiobitory effects of TNP-470 on the metastasis of hormone-irjdependent rat prostatic cancer AT 6.3 in vivo. MATERIALS AND METHODS: Hormone-independent rat prostatic carcinoma AT 6.3 cells (2 x 105 cells/mouse) were inoculated subctaneously into the right axillary region of the flank of male BALB/ c-nu/nu mice on day 0. Animals were given TNP-470 subctaneously at doses of 10 and 30 mg/kg three times per week from day 0. As a control, the same volume of the vehicle was given to the mice. Mice were sacrificed 3 weeks after implantation. Autopsy was performed, and tumors were resected immediately. The lung was processed for routine histological examination to detect metastases. RESULTS: TNP-470 inhibited pulmonary metastases in a dosedependent manner. The number of metastatic foci in the lung were 47.6+14.4 in the control group, 24.2+17.5 in the 10-mg/km group and 9.0+4.8 in the 30-mg/kg group. The widest diameters of metastatic foci in the lung were 0.55±0.08 mm, 0.31+0.14 mm and 0.15+0.1 mm in the control, 10-mg/kg and 30-mg/kg groups, respectively. CONCLUSION: TNP-470 exhibited a marked inhibitory effect on lung metastasis of the hormone-independent prostatic carcinoma. TNP-470 may be a useful antimetastatic drug for this tumor.

    A low carbon steel with 0.036 mass%C was deformed in tension at room and blue brittleness temperatures to determine the strains at which the micro-voids began to occur in the specimens at both temperatures. The natural strains determined were 0.30 at room temperature and 0.15 at blue brittleness temperature. It was also supposed that the dislocation density was about 2. 6 X 10° /cm2 in the specimens deformed up to the above strains.
