Veterinary Pathology 030098582110186-030098582110186 2021年5月31日Nestin, a class VI intermediate filament protein, is known to be expressed in various types of human neoplasms, including breast cancer, and is associated with their progression. However, its expression and role in canine mammary tumors remain unknown. We analyzed nestin expression in canine mammary tumors using in situ hybridization and immunohistochemistry. We also investigated its role in a canine mammary carcinoma cell line using RNA interference. Nestin expression was not observed in luminal epithelial cells of any of the 62 cases of benign mammary lesions examined, although myoepithelial cells showed its expression in most cases. In 16/50 (32%) primary mammary carcinomas and 6/15 (40%) metastases of mammary carcinomas, cytoplasmic nestin expression was detected in luminal epithelial cells. In luminal cells of primary mammary carcinomas, its expression was positively related to several pathological parameters that indicate high-grade malignancy, including histological grading ( P < .01), vascular/lymphatic invasion ( P < .01), Ki-67 index ( P < .01), and metastasis ( P < .05). Immunohistochemistry revealed that nestin expression was related to vimentin expression in mammary carcinomas ( P < .01). This relationship was confirmed using reverse transcription-quantitative polymerase chain reaction using 9 cell lines derived from canine mammary carcinoma ( P < .01). Finally, nestin knockdown in canine mammary carcinoma cells using small interfering RNA inhibited cell proliferation and migration based on WST-8, Boyden chamber, and cell-tracking assays. These findings suggest that nestin may at least partially mediate these behaviors of canine mammary carcinoma cells.

Japanese Journal of Veterinary Research 66(2) 63-70 2018年5月1日 査読有りWe investigated changes in cortisol (COR) concentration, which is well known as an index of stress in the serum of dairy cattle. The COR concentrations in serum obtained from dairy cattle were collected during practical training of first-year students on a farm attached to the Nippon Veterinary and Life Science University. Mean COR concentration in serum determined after practical training was significantly higher (P < 0.001) than that in serum collected before training. Discriminant analysis was used to classify the relation between COR concentration of serum collected before and after practical training. In conclusion, the data was bipartite according to the percentage of rise (rise rate) of COR concentration. Although the percentage of the rise was more than 300% in the high-rise-rate group, there was a significant negative correlation (P < 0.05) between age and COR concertation. It was thought that the high-rise-rate group has a chance to decrease stress after more experience. In contrast, the low-rise-rate group included 3 cattle indicating high COR concentration before and after practical training. Those 3 cattle were thought to be stressed easily. It is suggested that there was individual difference to stress.

Frontiers in Bioscience - Landmark 23(4) 614-625 2018年1月1日 査読有りWith the recent advent of whole genome and transcriptome sequencing technologies, long noncoding RNAs have been brought into the spotlight in molecular biology. H19 was one of the first reported long non-coding RNAs its expression is high in embryonic organs and absent or greatly reduced in most adult tissues. Accumulating evidence suggests that H19 plays crucial roles in embryogenesis. However, its levels are increased in different cancers, including breast, hepato-gastrointestinal, urological, respiratory, and brain tumors. Although there have been several controversial reports as to whether H19 is oncogenic or tumor-suppressive, most studies have indicated that H19 is associated with growth, migration, invasion, and/or metastasis in many cancers however, its reported functional mechanisms vary among cancer types. Furthermore, serum H19 levels in patients with certain cancers have been suggested to be useful for diagnosis and prognosis. Thus, H19 long non-coding RNA might be a candidate for development of promising therapeutic and diagnostic modalities for several cancers. The purpose of this review is to provide an inclusive report on the functional role of H19 in different cancers.

JOURNAL OF ZOO AND WILDLIFE MEDICINE 48(1) 265-268 2017年3月 査読有りA young adult, female, free-ranging Japanese raccoon dog (Nyctereutes procyonoides viverrinus) with scabies infection was found dead as a result of traumatic injuries presumed to reflect vehicular trauma. Necropsy showed a large solid mass located on the left ovarian region, occupying a third of the abdominal cavity. Histologically, the mass contained complex tissues derived from three germinal layers, with areas of cuboidal or columnar epithelium, keratinized squamous epithelium, bone, cartilage, and adipose tissue. This paper presents the first morphologic description of ovarian teratoma in a raccoon dog.

Primates 31(3) 427-429 1990年7月 査読有りSclerotic changes were found histologically in the myometrial vessels of 27 out of 33 cynomolgus monkeys (Macaca fascicularis). The sclerosis was composed of fibrous proliferations and intimal thickening. These changes were not observed in nulliparous cases, but were found only in multiparous ones. The findings suggest that the sclerotic changes in the myometrial vessels of cynomolgus monkeys are a pregnancy-induced phenomenon. © 1990 Japan Monkey Centre.

日本獣医生命科学大学研究報告 (65) 34-38 2016年12月1日A male, mixed breed, over 8 years old dog had a mass palpated in the right cryptochidism located at the inguinal subcutaneous region. At necropsy another large mass was observed in the retroperitoneum. The masses in the right testis and in the retroperitoneum were approximately 7.5 cm and 10.0 cm in diameter, respectively. Histopathological examination revealed that both masses were composed of polygonal cells with moderate to high grade atypia. The tumor cells formed alveolar or cord structures separated by fibrous stroma, and showed a tendency to orient perpendicularly to the basement membrane. Immunohistochemically, the tumor cells were positive for vimentin, inhibin-α and WT1. The testis was diagnosed with malignant Sertoli cell tumor, and the mass in the retroperitoneum was the metastasis from primary testicular tumor. In addition, marked hypoplasia of hematopoietic cells in the bone marrow, squamous metaplasia in the prostate and the gynecomastia were observed. It was suggested that the toxicity of hormones, such as estrogen and others, produced from the functional malignant Sertoli cell tumors was responsible for these lesions.
